Onset: Both Are Relatively Fast
Onset is one of the main similarities. Vardenafil is usually taken 25 to 60 minutes before sexual activity. Avanafil, sold as Spedra, is usually taken 15 to 30 minutes beforehand, and can begin working in as little as 15 minutes for some people.
Both work in the same way. They block an enzyme called PDE5, allowing blood vessels in the penis to relax and blood flow to increase during sexual stimulation. Neither causes an erection on its own.
Duration and Half-Life
Vardenafil's half-life is roughly 4 to 5 hours. Avanafil's is longer and more variable, generally cited as roughly 6 to 17 hours.
Half-life does not translate directly into how long a medicine provides a useful effect, so it should not be used on its own to predict duration for either one. Timing, individual response and tolerability matter more when choosing between them.
Why Their Doses Look So Different
Vardenafil comes in 5mg, 10mg and 20mg, with 10mg the usual starting dose. Avanafil comes in 50mg, 100mg and 200mg, with 100mg the usual starting dose.
Each medicine has its own independently established dosing scale, so the milligram numbers cannot be compared directly. A 100mg avanafil tablet is not ten times stronger than a 10mg vardenafil tablet.

Does Food Affect Either?
Both can be taken with food, but a high-fat meal can delay absorption. With vardenafil, the overall amount absorbed is not significantly changed. With avanafil, a high-fat meal can reduce its peak level and delay how quickly it starts working.
Is One Newer?
Yes. Avanafil reached the market considerably later than vardenafil. Being newer does not make a medicine better. It simply reflects when each was developed and licensed.

That depends on your priorities. Both are approved treatments for erectile dysfunction, and neither has been shown to be universally better.
