Mechanism of GLP-1: How GLP-1 Medicines Work (October 2026)

The mechanism of GLP-1 medicines is to copy a natural gut hormone that helps regulate appetite, slows how quickly the stomach empties and improves the way the body responds to insulin.1 Most people feel fuller sooner and less hungry between meals. Mounjaro (tirzepatide) also activates the GIP receptor.

A quick overview

What to know
before you start.

  • GLP-1 is a natural gut hormone; GLP-1 medicines activate its receptors to help regulate appetite.
  • They slow how quickly the stomach empties, so most people feel fuller sooner.
  • They also improve the way the body responds to insulin.
  • Tirzepatide (Mounjaro) activates both GIP and GLP-1 receptors, unlike semaglutide, which acts on GLP-1 alone.
  • These medicines work alongside a reduced-calorie diet and more physical activity, not instead of them.
In this guide

01 · The core answer

What is the mechanism of GLP-1?

GLP-1 medicines activate receptors for a natural gut hormone. Together with GIP, these hormone signals help regulate appetite, slow how quickly the stomach empties, and improve the way the body responds to insulin.1 In practice, most people feel fuller sooner and less hungry between meals, which supports a reduced-calorie diet.1

For a deeper look at the receptor biology, see our guide to the GLP-1 agonist mechanism, or the specific weight loss mechanism.

02 · What to do next

Is this relevant to me?

It is if you are an adult considering treatment for weight management. These medicines are prescription-only and used alongside a reduced-calorie diet and increased physical activity. If you are ready to explore treatment, our weight loss clinic explains the options, and the Mounjaro pen page shows current prices and the free online consultation.

03 · Single versus dual action

Single versus dual action

Tirzepatide is the first treatment of its kind to activate two natural gut-hormone receptors at once: GIP and GLP-1.1 This dual action is what distinguishes Mounjaro from single-action GLP-1 medicines such as semaglutide (Wegovy/Ozempic).1

Our guide to the tirzepatide GIP and GLP-1 mechanism goes into the dual receptor detail. If you want to compare, the Wegovy pen is the semaglutide option.

04 · What it means in practice

What the mechanism means in practice

The mechanism shows up as less hunger and earlier fullness, which makes a calorie-controlled diet easier to follow. In the SURMOUNT-1 trial of 2,539 adults, people taking tirzepatide alongside diet and exercise lost on average 15% of their body weight on 5mg, 19.5% on 10mg and 20.9% on 15mg over 72 weeks, compared with 3.1% with lifestyle changes alone.2

These are trial averages — individual results vary, and treatment works alongside a reduced-calorie diet and increased physical activity, not instead of them.2

05 · Related mechanism topics

Slower stomach emptying is part of why digestive effects can occur. A visual learner may prefer the mechanism animation guide. Newer tablet options are explained in how Foundayo works.

Tirzepatide stays in the body for a while after the last injection. It has a half-life of about five days, so after a final dose it takes roughly 4–5 weeks to clear the system — there is no way to speed this up, and nothing needs doing while it clears.1

How ordering works

How ordering works at EverydayMeds

Three steps, with our pharmacy team alongside you.

  1. EverydayMeds online consultation on a mobile phone

Complete a confidential health questionnaire covering BMI, medical history and current medication. No payment is taken until you're approved.

  2. A clinician ready to review an online consultation

Step 2: Prescriber review — usually same day

A UK registered prescriber checks your suitability against the licensed criteria (BMI ≥30, or ≥27 with a weight-related condition) and contraindications, and confirms your starting dose.

  3. An EverydayMeds delivery box

    Step 3: Next-day tracked delivery

Dispensed by our GPhC-registered pharmacy in temperature-controlled packaging with needles and a sharps bin included. Orders approved by 3pm for next-day tracked delivery.

Your questions, answered

Mounjaro price UK — frequently asked questions

Answers to common questions about Mounjaro prices, NHS access and ongoing treatment.

Ask our pharmacy team
What are the main GLP-1 drugs?
The best-known GLP-1-based medicines in the UK are tirzepatide (Mounjaro), semaglutide (Wegovy and Ozempic) and liraglutide (Saxenda). Mounjaro and Wegovy are once-weekly injections, while liraglutide is a daily injection. Mounjaro also activates the GIP receptor, so it is a dual-action medicine rather than GLP-1 alone.
How do GLP-1 medicines help people lose weight?
They help regulate appetite and slow how quickly the stomach empties, so most people feel fuller sooner and less hungry between meals. That supports eating fewer calories. They work alongside a reduced-calorie diet and increased physical activity, and individual results vary.
What is the downside to taking GLP-1?
Like any prescription medicine, GLP-1 treatments can cause side effects, and they are not suitable for everyone. The full list is in the patient information leaflet. A GPhC-registered prescriber reviews your suitability, and you can report any side effect through the Yellow Card scheme. Seek immediate medical help for severe, persistent stomach pain that may spread to your back, with or without vomiting (possible pancreatitis); pain in the upper right abdomen, fever or yellowing of the skin or eyes (possible gallbladder problems); or signs of a severe allergic reaction such as swelling of the face or throat and difficulty breathing.
Do I have to stay on GLP-1 forever?
There is no fixed answer; it depends on your goals, your health and your prescriber's advice. Tirzepatide clears the body over roughly four to five weeks after a final dose. Discuss any plan to stop or change treatment with your prescriber during your monthly reviews rather than deciding alone.
What organ is Ozempic hard on?
Ozempic is semaglutide licensed for type 2 diabetes only; it is not a UK weight-loss medicine. If you have kidney, liver or other health conditions, tell your prescriber before starting. Report any new or worrying symptoms to them and through the Yellow Card scheme.
What should I do before taking GLP-1?
Complete a consultation reviewed by a GPhC-registered prescriber, disclosing your medical history and every medicine you take. Read the patient information leaflet, and plan to use treatment alongside a reduced-calorie diet and increased physical activity. Our consultation is free, and support is available if you have questions.
