GLP-1 vs GLP-3: What's the Real Difference? (October 2026)

"GLP-3" is not an approved medicine class in the UK, so there is no licensed GLP-3 to compare with GLP-1. The term is usually used loosely for triple-receptor drugs such as retatrutide, which has no UK licence, or mistakenly for Mounjaro, which acts on two receptors (GIP and GLP-1).1 Licensed options today are Mounjaro and Wegovy.

For a UK adult wanting weight-loss treatment, the choice is between licensed medicines: Mounjaro or Wegovy. A "GLP-3" is not one of the options, because none is licensed or regulated for supply here. 02 · Is Mounjaro a GLP-3?

Is Mounjaro a GLP-3?

No. Tirzepatide activates two receptors, GIP and GLP-1, which together help regulate appetite, slow stomach emptying and improve insulin response.1 That dual action distinguishes it from single-action GLP-1 medicines such as semaglutide.

In the head-to-head SURMOUNT-5 trial, adults on tirzepatide lost on average 20.2% of body weight over 72 weeks versus 13.7% on semaglutide 2.4mg. Both are effective, licensed treatments.15

03 · Is Ozempic a GLP-3?

Is Ozempic a GLP-3?

No. Ozempic is semaglutide, a GLP-1 medicine licensed for type 2 diabetes, not for UK weight loss; private prescribing for weight loss is off-label. The licensed weight-management options are Mounjaro and Wegovy (semaglutide 2.4mg).18 A regulated UK pharmacy will not supply Ozempic for weight loss.

Retatrutide and other unlicensed drugs

Retatrutide has no UK licence. Mounjaro must not be combined with other GLP-1 medicines, unlicensed "research" peptides (such as retatrutide, which has no UK licence) or compounded copies, because these have no safety data and no regulated supply chain. Promises of faster results outside a licensed pathway are the warning sign. Report suspected side effects through the MHRA Yellow Card scheme. Only buy from a pharmacy you can verify on the GPhC register, never from social media, beauty salons or unverified websites. In SURMOUNT-4, people who stopped tirzepatide after 36 weeks regained on average 14% of body weight over the following year, while those who continued lost a further 5.5%. Stopping should always be planned with your prescriber, with diet and activity habits ready to carry the load.
What is known as the poor man's Ozempic?
Supplements such as berberine are marketed this way, but they are not medicines, are not regulated for this use and have no comparable trial evidence. Licensed options are prescription treatments assessed by a prescriber.
