What endogenous GLP-1 is and where it comes from
Endogenous GLP-1 is glucagon-like peptide-1 made inside your own body, as opposed to the synthetic versions given as medicines. 'Endogenous' simply means 'produced from within'. It belongs to a family of gut hormones called incretins, which are released when food passes through the digestive tract and which help the body handle what you have just eaten.
The main source is a group of specialised cells in the lining of the intestine, known as L-cells, found along the small and large bowel. Smaller amounts are made by certain nerve cells in the brainstem and by the pancreas. When nutrients such as carbohydrate, fat and protein reach these cells, they release the hormone into the bloodstream, where it travels to the pancreas, the stomach and the brain.
This is why the hormone is sometimes described as a conversation between gut and brain. After a meal, the signal tells the body that food has arrived. It supports the release of insulin when blood sugar rises, helps slow how quickly the stomach empties, and contributes to the feeling of fullness that makes you stop eating. Those effects are the same ones that GLP-1 medicines are designed to reproduce at a stronger and more sustained level.1
It helps to separate three ideas that online articles often blur. First, the natural hormone, which everyone makes and which rises and falls with meals. Second, the receptor it acts on, which sits on cells in the pancreas, gut and brain. Third, the medicines that bind to that same receptor. Mounjaro is licensed in UK adults for weight management and for type 2 diabetes, and it is a once-weekly pre-filled pen, not a hormone supplement.1
A final point of honesty: individual people differ in how much of the hormone they release after meals, and a lower natural response is not a character flaw or a diagnosis. Endogenous GLP-1 helps regulate after-meal blood sugar by supporting the body's insulin response when glucose rises. This is the meaning behind phrases such as 'endogenous GLP-1 regulates postprandial glycaemia': 'postprandial' means after eating, and 'glycaemia' means the level of sugar in the blood.
The process works in a few steps. You eat, nutrients reach the intestinal L-cells, and the hormone is released. In the pancreas it prompts insulin release when blood sugar is high, which helps move glucose out of the blood and into tissues. It also reduces the release of glucagon, a hormone that would otherwise raise blood sugar. Because the insulin effect depends on glucose being present, the system is self-limiting: the hormone does little when sugar is already low.
A second route matters just as much. The hormone slows how quickly the stomach empties, so glucose from a meal enters the blood more gradually instead of in a sudden surge. Mounjaro's dual action also helps slow how quickly the stomach empties and improves the way the body responds to insulin.1
A third route is appetite. Signals reaching the brain contribute to feeling full sooner, which is why many people on GLP-1 medicines describe feeling less hungry between meals. In practice, most people feel fuller sooner and less hungry between meals, which supports a reduced-calorie diet.1
In type 2 diabetes, this incretin system is part of why GLP-1 based medicines were developed. Mounjaro is licensed in the UK for type 2 diabetes as well as for weight management.1 If you have diabetes, any change to your treatment must be agreed with your diabetes team, and anyone taking insulin or sulfonylureas should know that doses may need adjusting because of the risk of low blood sugar.

06 · How long it lasts
How long endogenous GLP-1 lasts
Endogenous GLP-1 lasts only briefly: the body breaks it down very quickly, so its natural effect fades within a short time of being released.
The reason is an enzyme in the blood that cuts the hormone into an inactive form almost as soon as it appears. This design makes sense biologically. Eating is a repeated event, and a signal that vanished quickly allows the body to respond meal by meal without a permanent 'full' message. It also explains why the natural hormone alone cannot hold appetite down across a whole day.
That brevity is exactly what medicine design tries to overcome. Tirzepatide is built to stay active far longer. It has a half-life of about five days, which is why a single injection once a week is enough.1 The contrast with the natural hormone is dramatic: one acts briefly after a meal, the other provides steady receptor activation through the week.
The long half-life has practical consequences. After a final dose it takes roughly four to five weeks to clear the system, there is no way to speed this up, and nothing needs doing while it clears.1 This matters if you are planning a pregnancy, a surgical procedure or a change in treatment, because you should talk to a prescriber well in advance rather than assuming the effect stops the moment you skip a dose. Mounjaro must not be used during pregnancy, and it should be stopped at least one month before trying to conceive. If you become pregnant during treatment, stop the medicine and tell your prescriber promptly.
People sometimes ask whether 'making your own GLP-1 last longer' is possible through diet. A brief natural signal and a week-long medicine are different tools, and expectations should match.
07 · Boosting it naturally
Can you stimulate endogenous GLP-1 naturally?
The hormone is released when nutrients reach the gut, so eating a meal at all is the main natural trigger. Meals that include protein, healthy fats and fibre-rich plant foods are a sensible, balanced pattern, and they support fullness and steady blood sugar.
Supplements are the area where marketing runs furthest ahead of evidence. Products sold as 'GLP-1 boosters' or 'nature's Ozempic' are low-cost supplements with no clinical oversight. Dosing is not standardised, ingredient quality can vary, and dramatic weight-loss claims should be treated sceptically. They are not the same as a medicine licensed by the MHRA, and they are not a substitute for one. Ask a pharmacist before combining any supplement with prescription medicines, and report suspected side effects through the Yellow Card scheme.19
So can you get GLP-1 boosters in the UK? Supplements marketed that way can be bought without assessment, but they do not deliver GLP-1 and are not licensed to treat obesity. The only route to licensed GLP-1 receptor treatment is a prescription following a consultation.
What does help, regardless of hormones, is the foundation every licensed treatment is built on: a reduced-calorie diet and increased physical activity.1 Regular meals, adequate protein, plenty of vegetables and good sleep are unglamorous but reliable.
08 · Medicines compared
How GLP-1 medicines differ from the natural hormone
GLP-1 medicines differ from the natural hormone because they act for much longer and, in Mounjaro's case, on two receptors instead of one. Tirzepatide is the first treatment of its kind to activate two natural gut-hormone receptors at once: GIP and GLP-1.1
Together these help regulate appetite, slow how quickly the stomach empties, and improve the way the body responds to insulin. That dual action is what distinguishes Mounjaro from single-action GLP-1 medicines such as semaglutide (Wegovy and Ozempic).1 Mounjaro is a once-weekly injection supplied as a pre-filled multi-dose pen in six strengths: 2.5mg, 5mg, 7.5mg, 10mg, 12.5mg and 15mg.1
What does that mean in results? In the SURMOUNT-1 trial of 2,539 adults, people taking tirzepatide alongside diet and exercise lost on average 15% of their body weight on 5mg, 19.5% on 10mg and 20.9% on 15mg over 72 weeks, compared with 3.1% with lifestyle changes alone.2 These are trial averages, and individual results vary. Treatment works alongside a reduced-calorie diet and increased physical activity, not instead of them.2
To put that in practical terms, for someone starting at 100 kg, 20.9% is about 21 kg. That is a worked example of arithmetic, not a prediction for you, and trial averages do not predict your own result.
There is no comparable evidence for any food or supplement, which is the clearest way to see the gap between 'boosting your own GLP-1' and taking a licensed medicine.
Other GLP-1 based options exist. Wegovy (semaglutide) is a once-weekly injection with a dose ladder of 0.25mg, 0.5mg, 1mg, 1.7mg and 2.4mg, stepping up after at least four weeks to reach the 2.4mg maintenance dose from week 17.18 Liraglutide, sold as Saxenda, is supplied at 6mg/ml and is a daily injection rather than weekly. 09 · Safety
Which GLP-1 is safest for weight loss?
No single GLP-1 medicine can be named safest for everyone, because suitability depends on your health, your other medicines and your history. All licensed options have been assessed by the regulator, and a prescriber's job is to match the treatment to the person.
That is why a consultation matters. Counterfeit medicines are a genuine risk when GLP-1 treatments are sold outside regulated channels. Be wary of anyone offering to supply without a consultation or prescription, and check that a pharmacy appears on the GPhC register. Mounjaro is available on the NHS, but access is being phased in over several years. NICE recommends it for adults with a BMI of at least 35 plus at least one weight-related condition, and the NHS primary-care rollout began in June 2025 with the highest-need groups first, initially BMI 40+ with four or more qualifying conditions.46 Full rollout is expected to take years, which is why many people who meet the licensed criteria choose regulated private treatment rather than waiting.

11 · Storage and use
Storing and using a GLP-1 pen
Mounjaro pens are kept cold until first use: store them in the fridge at 2–8°C and they keep until the printed expiry date. This is the practical side of the contrast with your own hormone, which needs no storage at all.13
Once in use, a pen can be kept out of the fridge at a temperature not above 30°C for up to 30 days after first use, and must then be thrown away. Do not use a pen that has been above 30°C or has been frozen. Follow the pen's instructions for injecting, and use the guidance supplied with your order. If you are unsure about a pen that has been left out of the fridge before first use, check the patient information leaflet and contact the pharmacy rather than guessing.
Dosing starts low and rises gradually. After at least 4 weeks the dose can increase, depending on how you respond and tolerate it. Some people settle at a lower dose, and that is a normal outcome rather than a failure.
If you miss a dose, take it as soon as you can within 4 days (96 hours) of the scheduled time. If more than 4 days have passed, skip the missed dose and take the next one on your normal day — never take two doses within 3 days of each other.

Serious side effects: when to get help straight away
Serious side effects are uncommon but need urgent attention. Seek immediate medical help for: severe, persistent stomach pain that may spread to your back, with or without vomiting (possible pancreatitis); pain in the upper right abdomen, fever or yellowing of skin/eyes (possible gallbladder problems); or signs of a severe allergic reaction such as swelling of the face or throat and difficulty breathing. If you take insulin or a sulfonylurea alongside Mounjaro, watch for low blood sugar (shakiness, sweating, confusion) — your prescriber may need to adjust those medicines.1
