What "GLP-1" actually means

Tirzepatide is the first treatment of its kind to activate two gut-hormone receptors at once, GIP and GLP-1. Together these help regulate appetite, slow stomach emptying and improve the body's response to insulin. This dual action distinguishes Mounjaro from single-action GLP-1 medicines such as semaglutide.1

What the trials show

In SURMOUNT-1 (2,539 adults), tirzepatide with diet and exercise gave average losses of 15% on 5mg, 19.5% on 10mg and 20.9% on 15mg over 72 weeks, versus 3.1% with lifestyle changes alone.2 For someone starting at 100 kg, 20.9% is about 21 kg. In STEP 1, semaglutide 2.4mg gave 14.9% over 68 weeks versus 2.4% on placebo.9 SURMOUNT-5 is the first large head-to-head trial of the two. Individual results vary.

Other alternatives

Liraglutide products such as Saxenda are daily injections supplied at 6mg per millilitre, unlike once-weekly Mounjaro and Wegovy. Speak to a prescriber about regulated options instead.
What are the latest warnings about Mounjaro?
Only buy from a regulated pharmacy, as counterfeit medicines are a known risk; check the GPhC register. Contraception can be affected, since Mounjaro slows stomach emptying, which can reduce absorption of oral contraceptive pills. The SmPC advises switching to a non-oral method, or adding a barrier method (such as condoms), for 4 weeks after starting Mounjaro and for 4 weeks after each dose increase.1 Report side effects through the Yellow Card scheme.7 Seek immediate medical help for severe, persistent stomach pain that may spread to your back, with or without vomiting (possible pancreatitis); pain in the upper right abdomen, fever or yellowing of skin or eyes (possible gallbladder problems); or signs of a severe allergic reaction such as swelling of the face or throat and difficulty breathing.
